For single-step 96-well plate-based purification of genomic DNA from up to 50 mg of plant tissue (leaves, soft roots, seeds, fruit tissue, bud & blossom tissue)

The EchoLUTION Plant DNA Kit (96) has been developed to quickly purify genomic DNA from various species and types of tissues, including leaf, seed, fruit and root material (see table) with high throughput. No optimization of parameters like lysis time, temperature or elution volume is necessary.

The innovative EchoLUTION process in 96-well format does not need any washing steps and works completely in the absence of denaturing salts and organic solvents. These reagents - used in common Silica-based bind-wash-elute DNA extraction - inhibit enzymatic-reaction based analysis like PCR or NGS when carried over into the DNA preparation. The increase of overall speed, due to time savings from the process itself as well as the less error-prone enzymatic downstream analysis, makes EchoLUTION Plant DNA extraction kits the ideal tool for use in plant breeding projects.

Procedure

Duration:  Done in 60 minutes, including bead beating, tissue lysis and single-step column purification.

Typically the 96-well procedure starts with homogenization of the plant samples by bead beating in the lysis plate (Beating buffer provided, please order steel beads separately).  The subsequent lysis step takes place under physiological conditions where the enzyme activity of proteases is generally increased. BioEcho’s TurboLyse P Protease Mix (patent pending) contains a unique combination of several active enzymes with an expanded substrate specificity. All kinds of plant tissues can be lysed fast and successfully – simply and with the same protocol.

The EchoLUTION 96-well plate-based purification procedure combines an initial filtration, holding back potential cell debris, with an extremely efficient reverse purification within just one step. Following the lysis step, after addition of Clearing solution, the lysate is centrifuged, and the supernatant loaded onto the previously conditioned Purification Plate. The entire process is done after one additional centrifugation step. The purified high molecular weight genomic and mitochondrial DNA is eluted, while all debris and cellular compartments, inhibitors, small fragments (< 50 bp), and salts are completely removed. Since impurities are held back by the purification matrix and since the highly pure DNA is collected in the flow-through, the complete DNA is easily recovered with close to 100 % recovery and highest purity. Addition of carrier nucleic acids is not required since matrix interactions have been minimized.

Applications

For a molecular breeding project, various vegetables and fruits need to be purified in sufficient amounts with reliable high purity for later detection in qPCR or NGS. The EchoLUTION Plant DNA 96 workflow can be easily performed in high throughput.

The data set below, kindly provided by a Molecular Breeding Company (The Netherlands), show 8 different leaf tissues (4 samples each) from the indicated origin. Both OD-based purity indicators are in the optimal range. DNA is of consistent high purity and was reliably detected in qPCR.

Plant Breeding Project. 4 DNA preparations each purified with EchoLUTION Plant DNA 96 Kit from 8 different vegetable or fruit leaf punches were spectrophotometrically analyzed.
All samples were detected with high yield and optimal purity ratios (A260/280 and A260/230).

EchoLUTION-purified DNA comprises of particularly long DNA fragments, free of contaminants and enzyme inhibitors like chaotropic reagents and organic solvents and is highly suitable for all downstream applications such as PCR, Genotyping and NGS.
